1 What It Does

Semaglutide is the original blockbuster GLP-1 agonist — the most widely used and studied of the class. It slows gastric emptying and reduces appetite through GLP-1 signaling. The standard approach is a low 0.25 mg start with monthly step-ups.

The classic ladder: 0.25 mg/wk for 4 weeks, then step up (0.5 → 1.0 → 1.7 → 2.4 mg) every ~4 weeks as tolerated. Climb only as fast as side effects allow.

U-100 syringe: 100 units = 1 mL. At 3mg/2mL, 0.25 mg = ~17 units. Larger doses use more units — re-reconstitute a fresh vial with more water if draws get too large.
Other vial sizes

Semaglutide comes in 2–10 mg vials. As your dose climbs, a bigger vial with more water keeps draws manageable — recompute mg ÷ mL each time.
